Regulated nuclear import of Extradenticle (Exd) depends on a balance between nuclear export and localization signals. Shown here is the subcellular localization of NLS-myc-Exd in leg discs as detected by green staining with an anti-myc antibody. This construct contains an additional nuclear localization signal (NLS) derived from SV40 T antigen. The disc was costained for Homothorax (Hth; blue) and Dac (red), which serves as a marker for non-Hth-expressing cells. In the absence of Hth, NLS-myc-Exd was weakly nuclear.
