Abnormal mitotic spindle formation in Drosophila disc degenerate-4 (dd4) mutant cells. Shown here is the localization of centrosome associated protein CP190 with respect to spindle microtubules in a dd4 mutant cell at anaphase. dd4 encodes a 91 kD subunit of the Drosophila gamma tubulin ring complex. Immunostaining reveals that antibodies directed against CP190 (red) are weakly associated with the spindle poles, and accumulate in the vicinity of the mitotic chromosomes. The spindle microtubules are stained with antibodies directed against alpha -tubulin (green). In dd4 mutant cells, gamma -tubulin is dispersed throughout the cell. Thus, intact gamma -tubulin ring complex appears to be required at the poles in order to maintain the polar association of the CP190 protein. (For details, see Barbosa et al., p. 3126.)
