Dishevelled (Dsh) subcellular membrane localization is required for planar cell polarity signaling in Drosophila. Shown here are two immunoflorescence analyses of Dsh localization in pupal wings. Cells in the Drosophila pupal wing elaborate prehairs (stained red with rhodamine-phalloidin) that emerge from the distal vertex of each cell, and point distally. Planar cell polarity signaling controls the site of prehair emergence and the orientation of the prehair. Dishevelled (green in background image, white in foreground image) localizes to the distal (right) side of each cell, and directs the location of prehair initiation distally. Distal localization of Dishevelled is demonstrated by making clones of a Dishevelled-Green Fluorescent Protein transgene (foreground). (For details, see Axelrod, p. 1182.)
