Subnuclear sequestration of LEF1 by protein inhibitor of activated STAT-y (PIASy). Shown here are immunofluorescence analyses of COS7 cells that were transfected with expression constructs encoding LEF1 or T7-PIASy. LEF1 is distributed throughout the nucleus (green, upper left panel). PIASy, a nuclear matrix-associated SUMO E3 ligase, is localized in punctate subnuclear structures (red, lower left panel). Coexpression of LEF1 with PIASy results in a striking redistribution of LEF1 (green, center panel) into punctate structures that colocalize with PIASy (red, center panel). PIASy- and LEF1-containing nuclear bodies do not colocalize with heterochromatin (red, upper right panel), but partially colocalize with a subset of PML nuclear bodies (red, lower right panel), (For details, see Sachdev et al., p. 3088.)
