Expression of the transcription factor, Oct4, in the embryonic inner cell mass is required for pluripotency. Shown here are Oct4 expression analyses in cumulus cell mouse clones and subsequent outgrowths. Oct4-GFP is variably expressed in blastocyst-stage mouse clones (upper three panels). Quality of Oct4 expression in clone outgrowths, as visualized by in situ hybridization for Oct4 mRNA (background) or GFP expression (lower right panel), correlates with the ability to produce embryonic stem cell lines.
