Positioning the mitotic spindle relative to the cleavage apparatus in Saccharomyces cerevisiae. Shown here is a series of time lapse images of cytoplasmic microtubule capture and shrinkage at the yeast bud neck in cells expressing a GFP-TUB1 reporter construct to visualize astral microtubules (pink). To illustrate the dependence of microtubule capture at the bud neck upon septins, the fluorescent septin ring (yellow) from a cell containing a GFP-CDC3 construct was superimposed upon each image. This time sequence shows how one cytoplasmic microtubule (thin pink line) attaches to the septin ring (yellow), shrinks and pulls the spindle (pink) towards the bud neck, a crucial event for proper spindle positioning in yeast.
