Expression of an activated form of Drosophila Ras induces phosphoinositide 3-kinase (dPI3K) signaling in the developing wing. Shown here is an immunofluorescence analysis of a Drosophila imaginal wing disc using antibodies directed against the cell membrane marker CD2 (red). Clones of cells overexpressing dPI3K lack CD2 (red), and are enlarged due to increased rates of cellular growth. The levels and localization of phosphatidylinositol-3,4,5-triphosphate (PIP3), a second messenger generated by dPI3K, are visualized by a constitutively expressed reporter construct containing a pleckstrin homology domain fused to green fluorescent protein (GPH, green). GPH accumulates in the nucleus and cytoplasm of wild-type cells, but is recruited to the cell membrane of cells overexpressing dPI3K. (For details, see Prober and Edgar, p. 2286.)
