The vertebrate Ndc80 kinetochore complex is required for mitotic chromosome congression. Shown here is a prophase Xenopus tissue culture cell that was microinjected with function-blocking antibodies directed against the Xenopus homolog of Ndc80 (xNdc80) in the presence of the proteasome inhibitor MG132. The cell was subsequently fixed, treated with antibodies directed against tubulin (red), and stained with DAPI (blue) to visualize the DNA. Immunofluorescence analysis reveals that the unaligned chromosomes appear to float around a normal mitotic spindle and fail to congress to form a metaphase plate.
