The POU-domain transcription factors Oct-6 and Brn-2 are important regulators of Schwann cell differentiation and myelination in the developing peripheral nerve. Shown here is an immunofluorescence analysis of single nerve fibers derived from Oct-6-mutant mouse sciatic nerves at postnatal day 4, using antibodies directed against Brn-2 (green) and Neurofilament medium chain (red). Brn-2 is highly enriched in the nuclei of Schwann cells (green) as they ensheath axons (red). The activation of Brn-2 expression during Schwann cell development does not depend on Oct-6.
