Cover Cover The GATA-3 transcription factor is essential for cell lineage determination in the skin. Shown here is an immunofluorescence analysis of a GATA-3-null mouse whisker follicle at embryonic day 18.5. Antibodies directed against keratin K6 (green) of the outer root sheath's companion cell layer, and against hair keratins (red; monoclonal antibody AE13) of the hair shaft were used. GATA-3-deficient follicles fail to develop cells of the inner root sheath (IRS), which acts as a channel to guide the hair shaft through the skin surface. As a result, the companion cell layer (green) is abutted next to the hair shaft (red). Nuclei are stained with DAPI (blue). (For details, see Kaufman et al., p. 2108.)
