Cover Structural insight into inactivation of the retinoblastoma (Rb) tumor suppressor protein by the adenovirus E1A (Ad-E1A) oncoprotein. Shown here is the crystal structure of the pRb pocket domain (teal) in complex with the conserved region 1 (CR1) of Ad-E1A (red). The structure of the complex reveals that the Ad-E1A CR1 region and the E2F transactivation domain bind to the same site on pRb, thus explaining how Ad-E1A displaces E2F from pRb to mediate cell transformation by a mechanism that is distinct from its functional paralogs from other small DNA viruses.
