Cover A transgenic alternative splicing reporter system monitors the developmentally regulated switching of let-2 alternative splicing in Caenorhabditis elegans. Shown here is a fluorescent image of transgenic worms expressing a pair of splicing reporters that visualizes the developmental switching of the let-2 mutually exclusive exons—from exon 9 to exon 10—through the differential expression of green fluorescent protein (GFP) and red fluorescent protein (RFP). Embryos and early larvae express exon 9 (green), while adult worms express exon 10 (red). Using this system, color-based mutant screening was performed and alternative splicing defective-2 (asd-2) was identified as a regulator of the developmental switching of the let-2 mutually exclusive exons. Shown in the background is a large-scale image of the transgenic worms. (For details, see Ohno et al., p. 360.)
