Cover Maintenance of Drosophila Polo kinase expression is positively regulated by Myb and negatively regulated by E2F2–RBF. Shown here is an immunofluoresence analysis of a larval imaginal wing disc that is mosaic for a Myb-null mutant. The merged image displays GFP-tagged Polo (green); RFP-tagged HP1 (red) as an indirect marker for the presence of a wild-type Myb gene; and TOTO-3 (blue), a DNA dye with a preference for heterochromatin that was used to identify nuclei in the confocal plane. Cells lacking a Myb gene fail to maintain expression of GFP-Polo, even in very small mutant clones. (For details, see Wen et al., p. 601.)
